    摘要: 目的:觀察RNAi 抑制前列腺癌PC-3 細胞中的PHB 表達對細胞增殖、凋亡影響,探討PHB 對人前列腺癌的可能作用機制。方法:SiRNA 基因沉默PC-3 細胞中PHB 基因, Annexin V/PI 雙染和流式細胞計數觀察NM 和CDM 培養下及分別給予血小板源性生長因子PDGF 或EGF10 刺激的前列腺癌凋亡細胞數目差別 。結果:流式細胞儀(FCM)檢測發現,在siRNA 抑制PC-3細胞中的PHB 后,CDM 實驗組細胞凋亡率顯著增加,同NM 對照組比較有顯著性差異(p<0.05 );在CDM 組給予PDGF 或EGF 后,細胞凋亡率升高,與NM 對照組相比較有顯著性差異(p <0.05 )。結論:低膽固醇培養條件下前列腺癌細胞生長抑制,給予生長刺激因子促進細胞凋亡,低膽固醇抑制前列腺癌PC-3 細胞體外增殖活性可能同PHB 表達升高有關。

    25 Abstract: Objective:To observe the effects of RNAi inhibit PHB on cell growth,and apoptosis in Pc-3 cells.Explore the possible molecular mechanism of PHB on human prostate cancer.Methods:SiRNA knockdown PHB in PC-3 cells and treated cells with NM or CDM added/or not PDGF /EGF.Apoptosis was determined by Annexin V/PI double staining technique and flow cytometry.Results:Annexin V/PI double staining and flow cytometry discovered that after using siRNA knockdowned 30 PHB in PC-3 cells,PC-3 cells apoptosis rate of cultured in CDM was significantly higher than cells cultured in NM(p<0.05).Addred PDGF or EGF into CDM can upon apoptosis rate(p<0.05 ).Conclusions:Cholesterol depleted conditions can induce apoptosis in PC-3 cells.Cholesterol depleted conditions add growth factors can promote apoptosis in PC-3 cells.
